Prof. Brian M. Wolpin of Dana-Farber presented RASolute 302 at the ASCO Plenary Session: overall survival nearly doubled, and deterioration in pain and quality of life was significantly delayed.
On May 31, 2026, at the Plenary Session of the American Society of Clinical Oncology (ASCO) Annual Meeting, Prof. Brian M. Wolpin of the Dana-Farber Cancer Institute presented the full results of the Phase 3 RASolute 302 trial (abstract LBA5), with simultaneous publication in the New England Journal of Medicine.
Key results
- Median overall survival, overall population: 13.2 vs 6.7 months, HR 0.40;
- Median overall survival, RAS G12-mutant population: 13.2 vs 6.6 months, HR 0.40; one-year survival 53.3% vs 18.7%;
- Median progression-free survival, RAS G12-mutant population: 7.3 vs 3.5 months, HR 0.45; 6-month progression-free rate 58.7% vs 31.7%;
- Time to pain deterioration: 9.2 vs 3.8 months, HR 0.51; time to deterioration in global health status/quality of life: 5.7 vs 2.6 months, HR 0.60.
Safety
Grade ≥3 treatment-related adverse events 43.6% vs 57.5%; dose reductions 36.1% vs 57.5%; discontinuations due to adverse events 1.2% vs 11.2%. Rash (17.4%) and stomatitis (6.6%) were the adverse events most often leading to dose reduction.
The discussant described the results as a "transformative moment" in the treatment of pancreatic cancer.
